Selank is a synthetic analog of the endogenous peptide tuftsin (Thr-Lys-Pro-Arg) with an additional Gly-Glu-Thr sequence that extends its half-life and enhances CNS penetration. Developed at the Institute of Molecular Genetics of the Russian Academy of Sciences, Selank has been clinically used for generalized anxiety disorder, phobias, and cognitive enhancement in Russia since 2009.
The GABAergic Mechanism
Selank's anxiolytic effects are mediated primarily through modulation of the GABAergic system — the same system targeted by benzodiazepines. However, unlike benzodiazepines, Selank does not cause sedation, cognitive impairment, tolerance, or dependence. It acts as a positive allosteric modulator of GABA-A receptors in a subtype-selective manner that produces anxiolysis without the cognitive side effects associated with non-selective GABA-A modulation.
Cognitive Enhancement: The Paradox
Most anxiolytic compounds impair cognition as a side effect of their sedative mechanisms. Selank is unique in that it simultaneously reduces anxiety and enhances cognitive performance — a combination that has made it particularly valuable for high-performance individuals dealing with anxiety-driven cognitive impairment. Clinical studies have demonstrated improvements in attention, working memory, and processing speed alongside anxiety reduction.
BDNF and Neuroplasticity
Like Semax, Selank also upregulates BDNF expression, suggesting that its cognitive benefits extend beyond simple anxiolysis to include genuine neuroplasticity enhancement. The combination of anxiety reduction and BDNF upregulation makes Selank particularly effective for individuals whose cognitive performance is limited by anxiety-driven stress responses.
